Guidelines for determining the amount of volume recovered from the LeviCell 1.0 cartridges.
Each run in a LeviCell 1.0 cartridge requires 220ul of Levitation Agent in buffer. Users can expect to recover 60 - 80% of the total volume from the top and bottom output wells combined. There two way to measure your volume recovered in the output wells:
1. Estimate with a pipette. After the user has recovered their sample into a plate or tube, they should set their pipette to 100ul (the recovery volume will only be >100ul if the split line is set to +/-10) and aspirate the sample. Slowly adjust the volume dial down until the liquid reaches the pipette tip. This is the estimated volume recovered.
2. Estimate using an analytical balance. To do this, weigh empty tubes (1.5 or 2ml) for the output wells using an analytical balance with units set to grams with 4 significant digits. Once levitation is done, recover the sample into the same pre-weighed tubes and weigh again. The difference in the "full" tube and the empty tube is equivalent to the number of mls in the tube. Multiply that number by 1000 to determine the total number of ul recovered. Always allow the balance to stabilize before recording values. See the graphic below for instructions.
